CogRx: Hope on the Horizon

Macro-molecular structures brought to life

This is an animation created for CogRx, a small biotech startup in Pittsburgh. This was an exciting project that brought to life the protein structures CogRx is currently investigating. For many structural biologists, the proteins they study are only glimpsed through test results, or piece-mail through visualization software. Our visualization scientists specialize at building complex macro-molecular structures. We work with structural biologists to then determine protein behaviors and interactions based on different surface properties, binding and active site locations.

Aligning Known Structures to Target Proteins


Some proteins that we know interact have yet to be crystallized as bound structures. In order to better understand protein function and the cascade of events that result in cellular processes, we work with structural biologists to theorize potential alignments and interactions. In the slides above, our biomedical visualization scientists were required to identify the binding location and orientation of a HEME group to the functional residues of PGRMC1. To do this, we conducted a comparative analysis of unbound PGRMC1 to a receptor analog bound by a HEME group. Through structural alignment of functional residues, we not only correctly placed the HEME group, but in doing so identified unique structural dynamics of PGRMC1 not previously known. This theory was later confirmed through experimental test results.

Determining protein dynamics through data analysis

Creating the 3D animation for the beta amyloid beta oligomer required significant research in order to determine viable conformation changes of individual monomers. Referencing Dr. Vijay Pande’s work at Stanford University to simulate AB oligomerization, our medical animators developed this depiction of an AB oligomer movement.
